Stone formation in the urinary tract has occurred for thousands of years. Archaeologists have found several cases of urinary tract calculi in Egyptian mummies, dating back 7000 years ago.143 , 147 Hippocrates in 387 bc documented the association of urinary tract infection and urolithiasis. Approximately 2000 years later, Marcet noted the combination of phosphate calculi with alkaline, ammoniacal urine, and putrefecation.53
In 1901, Brown15 proposed the theory of bacterial splitting of urea as a promoter of stone formation. His six patients' stone analyses revealed “phosphates and carbonates of calcium and magnesium.” All six patients had Proteus vulgaris isolated from urine cultures.15 A year later, Hagar and Magath54 suggested that a bacterial protein “urease” was responsible for the hydrolysis of urea. Sumner131 isolated urease from the jack bean, Canavalia ensiformis, and was later awarded the Nobel Prize for this work.
