Highlights

Novel applications of double divisor-ratio spectra derivative spectrophotometry and difference spectrophotometry as green techniques for resolution of mixtures.
This proposed method could be considered superior, science it is simple inexpensive sensitive and rapid taking into account that the analysis of aliphatic compounds is challenging.
The proposed technique was effectively applied for estimation of the four investigated drugs in their commercial preparations, where the results are compared to previously reported methods.
The greenness profile of the proposed methods was evaluated utilizing the most recent greenness assessment tool (AGREE).

Summary

Background

Green spectrophotometric methods were developed and validated for determination of some CNS active drugs as antiepileptics and brain stimulants.

Objective

Piracetam (PIR), Levetiracetam (LEV) and Brivaracetam (BRV) were assayed as a ternary mixture using double divisor-ratio spectra derivative (DDRSD) (method I). One more binary co-formulated mixture of Piracetam (PIR) and Vincamine (VIN) was assayed using difference spectrophotometric procedures (method II).

Method

I was applied to determine PIR at 302nm in the first derivative of the ratio spectra in the selected spectral region. The content of LEV was determined by measuring the spectra at 215nm in the first derivative of the ratio spectra in the selected spectral region. The concentration of BRV was estimated by measuring the first derivative of the ratio spectra in the chosen spectral region and detecting the signals at 229.7nm. The application of method (II) procedures resulted in measuring the absorbance of PIR at 220nm which is the zero crossing point on the difference spectra of VIN in 0.1M NaOH vs. 0.1M HCl. Similarly, the absorbance of VIN was measured at 245.0nm, which is the zero crossing point on the difference spectra of PIR.

Results

The suggested methods were fully validated adopting ICH guidelines. The linearity ranged from 10-100μg/mL for the three racetams and from 2–20 for VIN. The recovery percentages were ranged from 98.72% to 101.8% for method I and from 98.13% to 101.06% for method II. Moreover, the proposed methods were proved environmentally benign using the most recent assessment tool named AGREE.

Conclusion

Both procedures were successfully applied for the determination of each drug in bulk powder, checked using laboratory prepared mixtures, and directly applied on commercially available pharmaceutical products without interference. The obtained results revealed a good agreement with those obtained by the reported methods.

Contexte

Des méthodes spectrophotométriques vertes ont été développées et validées pour la détermination de certains médicaments actifs sur le SNC comme antiépileptiques et stimulants cérébraux.

Objectif

Le piracétam (PIR), le lévétiracétam (LEV) et le brivaracétam (BRV) ont été dosés en tant que mélange ternaire à l’aide d’un dérivé de spectres à double diviseur (DDRSD) (méthode I). Un autre mélange co-formulé binaire de Piracétam (PIR) et de Vincamine (VIN) a été dosé en utilisant des procédures spectrophotométriques différentielles (méthode II).

Méthode

La méthode I a été appliquée pour déterminer le PIR à 302nm dans la première dérivée des spectres de rapport dans la région spectrale sélectionnée. La teneur en LEV a été déterminée en mesurant les spectres à 215nm dans la dérivée première des spectres de rapport dans la région spectrale sélectionnée. La concentration de BRV a été estimée en mesurant la première dérivée des spectres de rapport dans la région spectrale choisie et en détectant les signaux à 229,7nm. L’application des procédures de la méthode (II) a abouti à la mesure de l’absorbance de PIR à 220nm qui est le point de croisement zéro sur les spectres de différence de VIN dans NaOH 0,1M vs HCl 0,1M. De même, l’absorbance de VIN a été mesurée à 245,0nm, qui est le point de passage par zéro sur les spectres de différence de PIR.

Résultats

Les méthodes suggérées ont été entièrement validées en adoptant les directives de l’ICH. La linéarité variait de 10 à 100μg/mL pour les trois racétams et de 2 à 20 pour le VIN. Les pourcentages de récupération variaient de 98,72 à 101,8 % pour la méthode I et de 98,13 à 101,06 % pour la méthode II. De plus, les méthodes proposées ont été prouvées sans danger pour l’environnement à l’aide de l’outil d’évaluation le plus récent appelé AGREE.

Conclusion

Les deux procédures ont été appliquées avec succès pour la détermination de chaque médicament en poudre en vrac, vérifiées à l’aide de mélanges préparés en laboratoire et appliquées directement sur des produits pharmaceutiques disponibles dans le commerce sans interférence. Les résultats obtenus ont révélé un bon accord avec ceux obtenus par les méthodes rapportées.
