Evaluation of efficacy of aloin in treating acute trauma in vitro and in vivo - 13/03/17

Abstract |
Clinical multiple acute trauma has brought great suffering for patients. The use of traditional Chinese medicine provides a new approach to treating trauma. Aloe Vera gel has been used to treat skin diseases for a long time, but its efficacy is not stable. As the most important part of the organic active component of Aloe Vera gel, the effect of aloin in acute trauma has not been reported. In this article, the effect of aloin on promoting wound healing was investigated through a series of experiments in vitro and vivo. For the in vitro experiment, 3-(4,5 -dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cell proliferation and cycle analysis demonstrated that aloin could enhance the proliferation of human skin fibroblasts and endothelial cells. Transwell and scratch tests demonstrated that aloin could increase the chemotaxis and migration on human skin fibroblasts, and the tube formation assay demonstrated that aloin solution significantly increased the tube network formation compared to the control group. For in vivo experiments, we used a rat model of acute trauma to illustrate that aloin solution significantly accelerated wound healing. Moreover, the results showed that aloin can accelerate wound healing by inducing the expression of epidermal growth factor, and additional, new neovascularization and fibroblasts were found in the skin treated with aloin solution. Additionally, collagen fibers are arranged in a neat and orderly arrangement compared with the control group. Aloin solution could promote the rate of wound closure, and the collagen fiber arrangement of the healed skin was more regular, which inhibited the formation of scar tissue.
